They are referring back to the eyes which is the imprint for Aita. We haven't seen a case with both the eyes and the neck mark. Who would she even be if she had the neck mark? Everyone that used that method is accounted for.

As someone with partial heterochromia, thank you for this, Lacrosse. I'm so tired of always being accused of being Isu/sage, this witch hunt has to stop.😩

I think this is meant to demonstrate the Order's brutality and thoroughness when purusing the power of the Isu. They dissected a teenage girl just because she had this rare condition that Sages also have.
